Publication Quality DNA Sequencing Service

Sequetech DNA Sequencing Our Publication Quality Sequencing Service is offered for Plasmids, PCR Products, and larger constructs such as Lambda and BAC clones.  Sequencing reactions are performed; primers are designed and synthesized to allow us to "Primer Walk" the region of interest.  Both strands of the region of interest are sequenced independently.  The accuracy of Publication Quality sequence data is guaranteed by virtue of complete complementarity between the two strands. Project turnaround is dependent upon the size of the region to be sequenced and pre-existing knowledge of the expected sequence. Sequencing is typically completed at a rate of about 1.5 Kb to 2.0 Kb per week.



Here is what is included in this service:

Analysis of sequencing reactions on ABI 3730xl DNA Analyzer platform
Each base is independently confirmed on both strands through redundant sequencing analyses.  
Overlapping sequencing analyses performed on each strand.  
Assembly of overlapping sequence data
Design and synthesis of all necessary sequencing primers
Full support for templates which prove to be difficult to sequence
An electronic text file of the finished sequence
Quantifying template concentration
An optional report which includes:
         a printed copy of the entire text sequence
         a translation of the finished sequence in all 6 reading frames
         a map indicating the sequence data coverage and sequencing primer locations
         a printed copy of the four-color electropherograms from each sequencing run
         a list of the sequencing primers with their sequences
         a map of the cut sites of all commercially available restriction enzymes

Templates must be purified prior to submission.  Exception: If you are also requesting our RCA Service, you needn't purify your templates.

The quality of sequencing results is highly dependent on the quality of the DNA.  Good results have been obtained from both cesium chloride purified and Qiagen column purified plasmids.
